A template website is basically when your site is designed using a specific template. This can vary in degree from a very strict template that you have very few elements you can change to one where the layout is set but you can add many elements within the layout.

Bespoke is exactly what is says, where you can pretty much create a site from scratch adding areas and functionality where you want. A bespoke website is most likely to cost you more but gives you a lot more flexibility. You should check if you are going to use a template site, how much you can change and if there are many other websites out there using exactly the same one, as really you want to be able to stand out from the crowd a bit.
